Special remarks: Neuropathologically Alzheimer's disease (AD) is characterized by widespread deposition of amyloid beta protein (Abeta) in the cerebral cortex, and since the intracerebral accumulation of Abeta is highly specific for AD and the gene mutation responsible for familial amyloidosis causes Abeta to increase, Abeta is thought to be of extremely great significance in the pathogenetic mechanism of AD. Abeta is generated by proteolysis of the Abeta precursor protein (APP), a type I membrane protein. The enzyme that cleaves it at the N-terminal is called beta secretase, and it is regarded as being the protease that acts at the initial step in the production of Abeta. The major beta secretase has been identified as beta-site APP Cleaving Enzyme 1 (BACE1), and BACE2 has been isolated as its homologue. Since increased BACE1 activity has also been reported in the brain of patients with sporadic AD, BACE1 inhibitors have been attracting attention as therapeutic agents for AD.
The kit can apply to assay of human, mouse and rat BACE1.
